Even if they are separated, the operation is complex and the success rate is not high. To this end, we will introduce a method for the isolation of bacteria - the pleating method. Select the first mature mushroom that has been unfolded in the cap. Cut about 1.5 cm of the pleat at 2/5 of the front of the pleat. Pick a piece of pleat with tweezers and attach it to the sterilized bevel. Immediately above the culture medium, the test tube is then placed upright in a beaker or jar so that the spores can project freely onto the medium. When the surface of the culture medium is seen to have spore imprints, the bacteria pleats are taken out of the flame of the alcohol lamp in the inoculation box, and the test tube is put into an environment of about 25° C.
